Though you are doing diploma, your highest qualification with all the three subjects i. e physics chemistry and Biology remains 12th senior secondary. And also, it doesn't matter even if it's your 2nd attempt in neet.
While, those who are doing Bsc and have all the three subject i. e physics chemistry and Biology needs to select code 5 and those who have completed BSc with at least any two of physics chemistry and Biology in their bsc need to select code 6.

The cutoff of NIPER JEE varies each year according to the number of candidates, number of seats, difficulty level of the question paper, etc. In the year 2019, the NIPER JEE cutoff rank was 706 for General category and 711 for reserved categories (OBC, SC, ST and other reserved categories). As per their ranks and categories, candidates will be allotted to different branches of NIPER all over India. Whereas, candidates with higher ranks (roughly around 100 for unreserved and 300 for reserved categories) will be able to bag seats in more distinguished branches.

Admission in mbbs in through neet examination.
It doesn't matter even if you had failed in your class 12th board examination in 2019 . Now that you have passed your class 12th in 2021 you are eligible for Neet examination provided you must have passed it with physics, chemistry ,biology/biotechnology and English and must have got:-
at least 50 % in aggregate of PCB i.e Physics, Chemistry and Biology/Biotechnology in 12th examination if belongs to general category
at least 40% in aggregate of PCB if belongs to Obc/ Sc/St
at least 45 % in aggregate of PCB in case of pwd candidates to be eligible.
So, if you fulfill this criteria then you are eligible for Neet examination and after qualifying neet you can get admission in MBBS as well on the basis of your rank obtained in neet
Also,
* to be eligible for neet 2021 one has to be at least 17 years old or more by 31st December 2021
* There is no upper age limit for Neet 2021
* There's no limit for the maximum number of attempts/times one can appear in NEET.

Yes, definitely. You can give 12th public exam whenever you want. Failing in an exam does not mean that you cannot give it again. You can prepare for the exams with dedication again and can pass them with flying colors. Some private institutions have their own age regulations which allows only students below 17 year old to give exam but this is not a case in every institution. Also, there is an option of open boards to give board exams where there is no age limits.

Students who wish to take admission in 10th class of Rajasthan secondary Board shall have to to reach the age of 14 years but not exeeding this age limit and have qualified up to 9 class standard from Rajasthan secondary Education Board or any other recognised Education Board. They have to fulfill the criteria of 10th class admission under Rajasthan secondary Education Board.(RSEB).Rajasthan secondary Education Board takes the exam of class 8 , 10 and 12.

The number of attempts(years) for JEE Main is three. And these are like this, first one is when you are about to pass the class 12th or just passed. And two more attempts in the next two succeding years.
Now for JEE Advanced, you get one less attempt than JEE Main. So, the total number of attempts(years) for JEE Advanced is two. One in the passing year of JEE Advanced and the other one is in succeding year.
Now coming back to your question, see if your brother is giving improvement exam in 2022 and his original 12th exam is in 2021 then he will get 3 chances for JEE Main in the year 2021, 2022 and 2023, but if he is freshly giving 12th exam in 2022 I mean not giving improvement then he will get three chances and these are in 2022, 2023 and 2024.
You can think the same for JEE Advanced but you will get one less year of attempt.
