No .. NPTEL exam score will not effect your semester Grades . So , if you can't perform well in NPTEL exam , don't worry .. you'll not get any Backlog .
Generally NPTEL exam is given by students of Technical colleges for MAR marks (Mandatory Additional Requirement ).. so you can see, that it's written additional in the term MAR , not compulsory .
and these type of online courses basically comes under MOOCs course . MOOCs course is necessary for college students just for extra credit points in MAR .

NPTEL certification in programing will help you to up-skill your Programming knowledge and can help you to get better job opportunities when compared to others who did not have those certifications through NPTEL. But it alone not sufficient to get job in IT sector. It is necessary to have Undergraduation degree to get good job in IT sector and just with NTPEL certification it is not that easy. Better to do Undergraduation degree like BCA, B.Sc Computer Science etc for better job opportunities.

NPTEL generally happens twice a year once during the summer and the other during the winter. If you have failed this time you will not be given a certificate for the subject, however you will be able to take it up again in the next season. The choice is given to you, you can either take up the same subject or you can opt for another subject as well.
