Yes, you will be able to appear for JEE Main examination if you have completed a diploma recognized by AICTE or a state board of technical education of at least 3-year duration.
However, you can only attempt the exam for the purpose of qualifying to appear for JEE Advanced. Your rank and score won't be considered for admission in the NITs and CFTIs, diploma candidates are not eligible for these admissions.

Yes, Diploma students are eligible for JEE mains. But you won't be eligible for admission into NITs and other GFTIs. You can appear for JEE Mains Qualify it to appear for JEE Advance. If you qualify JEE Advance you can get admission into first year of Engineering Courses of IITs.
Diploma Students can also give other lateral entrance exam to get admission into second year of engineering in various colleges
