No it is not necessary or compulsory to take the April session JEE Mains exam if you've attempted the January session of JEE Mains. You have the option to attempt the April session if you are not satisfied with your performance in January session or if you wish to give it another chance and perform better.
In the end while computation of the final merit rank list, the best of the two scores will be considered. Hence the session in which you score higher from January or april will be considered to compute your final rank.
So you can attempt the april session if you wish to give it another shot for improving your score. And the one in which you score higher will be considered.

No ,It is not necessary to give the 2nd phase of jee mains of exam.It 's on you if you want then you can give 2nd phase exam .Best of marks from these 2 phase of exam is considered as raw marks.If you don't give 2nd phase of mains exam then your score of 1st phase exam is considered as your best score.
