Glucagon, in conjunction with the hormone insulin, helps to regulate blood sugar levels and keep them stable. Insulin is released to prevent blood sugar levels from falling too low (hypoglycemia), whereas glucagon is released to prevent blood sugar levels from rising too high (hyperglycemia) (hyperglycaemia).

Low blood glucose, protein-rich meals, and adrenaline all stimulate the production of glucagon (another important hormone for combating low glucose). The release of glucagon is inhibited by elevated blood glucose and carbohydrate in meals, which are sensed by pancreatic cells.

In the long run, glucagon is essential for the body's response to hunger. For example, in order to save the limited supply of glucose, it encourages the usage of stored fat for energy.
