According to the guidelines of UGC, a Professor need to put 20 hours of Teaching which also includes practicals. So they have working hours from morning 9 to till 5 in the evening. Here they need to work for 40 hours in Overall for a week.

The working hours of requirement for any professor in a private autonomous degree College depends on the rules and policies adopted by that particular college in individual.
But as per the UGC guidelines,a working professional or a teaching assistant needs to work at least 20 hours a week including his practicals and 40 hours in his overall duration to make his work Tally count.

UGC NET exam is conducted for Eligibility of Assistant Professor or Junior Research fellow . UGC NET will be conducted by NTA.
Candidates can apply for this exam after completing Master's degree from any recognized Institute. Also, they need to score 55% marks in the Master's degree (5% relaxation for reserved category). In your case, you are eligible to appear for UGC NET if you have cleared the above mentioned eligibility criteria.

Well usually the working hours are like normal that is from 9 to 5 in the evening but since most of the professors have their post doctoral work going on and research work going on, they might stay in the college or lab for as long as they wish and have work to do. But official hours usually is between 9 to 5.
