JEE main is an entrance exam which provide admission for B.E and btech courses in different universities all over india.
There is no difference between JEE mains and JEE advanced except for the fact that JEE advanced is a lot tougher than JEE mains and JEE advanced is for admission in indian Institutes of technologies (IITs).
No matter how good you score in mains without qualifying JEE advanced you cannot get IITs. For this year the 75% in class 12 in JEE advanced criteria has been relaxed. To seat for jee advanced you have to qualify JEE mains.
JEE mains/ advanced has 3 subjects namely physics, chemistry and maths and you are assessed on this 3 subjects only. The topics you must focus on are:
Maths: Trigonometry, integral calculus, differential calculus,Vectors, analytical Geometry, algebra
Physics: general physics, thermodynamics, optics,modern physics, mechanics,electricity, magnetism, kinematics
Chemistry: physical chemistry, organic chemistry, Inorganic Chemistry, electrochemistry, mechanisms of organic reactions

There are only three subjects in JEE Mains and Advance syllabus. And all the three subject takes equal importance in JEE Mains and Advance. In JEE Advance every subject need to complete cut off marks to get selected. But I can say chemistry is the subject which can help you to improve your score, because in chemistry there are less chance of question to be wrong as every question is concept based.
