is maths necessary in 12th to apply for iiser?

No it is not, most of the students have PCB group in 12th standard and they can also apply for IISER by avalible channels.

If you choose biological sciences then not necessary

If you are applying for physics,chemistry or mathematics, then you need math. For biological sciences it's not required.
